Inhibitory Mechanism of Reduced Glutathione against the Activities of Cholera Toxin



Abstract
Biologically important activities of cholera toxin are enterotoxic activity, receptor binding property and antibody fixing properties. This study is of the detailed elucidation of inhibitory mechanism of reduced glutathione against the activities
of
cholera toxin. cholera toxin. was treated with the reduced glutathione, and then evaluated the enterotoxic activity, receptor binding property, antibody fixing property, eletrophoretic pattern and Ellman assay for thiols. Cholera toxin treated
with
oxidized glutathione was used as control.
@ES the results obtained were as follows.
@EN 1. Enterotoxic activity of reduced glutathione treated cholera toxin was significantly inhibited (p<0.05). In rabbit vasopermeability factor assay, Blueing(Induration) was measured 47§±¡¾30(134§±¡¾52) in mean diameter, compared with Control
of
88§±¡¾34(208§±¡¾34).
2. In combined CM1-and antibody-ELISA for evaluating both the receptor binding activity and antibody fixing properties, Reduced glutathione treated cholera toxin did not show any meaningful difference in comparison with control.
3. In native electrophoresis, Both reduced glutathion treated cholera toxin and control exhibited the same patten of protein bands.
4. In Ellman assay for estimating thiols quantitatively, Reduced glutathione treated cholera toxin contained more thiols(O.D. 0.09¡¾0.02) than control(0.05¡¾0.01)(p<0.05),
These results indicate that inhibitory effect of the reduced glutathione against the activity of cholera come about the direct and not-active thiol-disulfide exchange between cholera toxin and reduced glutathione, and reduced glutathione did not
affect
the receptor and antibody binding activity of cholera toxin.
